PACE PCR genotyping

PACE® 技術原理

PACE® 技術原理與 KASP™ 相似,是利用兩條競爭性等位基因專一性引子 (competing allele-specific primers) 與通用螢光報導系統來區分不同的基因型。一個標準的 PACE® 反應主要由 DNA 樣本Assay Mix(引子組)Master Mix(預混液)三個部分組成,其中:

引子組 (Assay Mix):包含三種未標記的 primers,分別是兩種等位基因專一性前置引子 (allele-specific forward primers),用於辨識目標變異位點。兩者序列僅在 3' 末端鹼基不同(對應兩種不同的等位基因),且各自帶有一段與 FAMHEX 螢光報導探針互補的 5' 端尾巴序列 (tail sequence);一種共同反置引子 (common reverse primer),結合於目標序列的反股上。

預混液 (Master Mix):包含 PCR 擴增所需的基本元件,以及通用的 FRET 螢光報導匣 (universal FRET cassettes)。報導匣含有兩種 FRET 螢光探針,分別帶有 FAMHEX 螢光染料;在未與互補序列結合的狀態下,螢光訊號會被探針結構上的淬滅劑 (quencher) 所抑制。與目標序列完全匹配的等位基因專一性引子,在 PCR 反應中具有較高的延伸效率,因此於擴增過程中逐漸取得競爭優勢。隨著 PCR 進行,專一性引子的 5' 尾巴會被整合到新合成的 DNA 股中。預混液中的螢光報導匣會與剛剛產生的尾巴互補序列結合,隨著新的延伸反應,螢光染料會與淬滅劑分離,釋放出螢光訊號。藉由判讀螢光訊號,即可獲知樣本的變異位點是屬於同型合子 (homozygous) 還是異型合子 (heterozygous)。
