小鼠脂多醣 (lipopolysaccharide, LPS) ELISA 試劑盒採用三明治型酵素免疫分析法 (Sandwich ELISA)，可定量分析細胞培養上清液、血清、血漿及其他生物體液樣本中的小鼠 LPS 濃度。檢測時，先將標準品或樣本加入試劑盒提供的微量盤中，盤孔底部已預先包被 LPS 專一性捕捉抗體。接著加入帶有辣根過氧化物酶 (horseradish peroxidase, HRP) 標記之 LPS 專一性偵測抗體，形成抗體－抗原－抗體三明治複合體。沖洗去除未結合物質後，加入 HRP 受質溶液 (substrate solution) 進行呈色反應，最後加入終止液 (stop solution) 停止反應，並於 450 nm 測定吸光值。在檢測範圍內，450 nm 吸光值與樣本中的 LPS 濃度呈正比，可透過標準曲線定量分析 LPS 濃度。

檢測原理

三明治 ELISA (Sandwich ELISA) 是檢測蛋白質、生長因子及細胞激素等生物分子的常用免疫分析技術，具有高專一性與高靈敏度。該方法採用兩株辨識不同抗原表位的專一性抗體，分別作為捕捉抗體與偵測抗體（偵測抗體通常帶有 HRP 酵素標記）。當樣本中含有分析物時，可形成「捕捉抗體－分析物－偵測抗體」三明治複合體，加入 HRP 呈色受質後產生顯色反應，其顏色深淺與樣本中的分析物濃度呈正比，透過標準曲線即可定量分析樣本中的分析物濃度。

科學背景

脂多醣 (lipopolysaccharide, LPS) 是革蘭氏陰性菌 (Gram-negative bacteria) 外膜的重要組成成分，具有內毒素 (endotoxin) 特性，是研究先天免疫反應與發炎反應的重要刺激因子。LPS 可被脂多醣結合蛋白 (LPS-binding protein, LBP) 捕捉並傳遞至 CD14，進一步與 Toll-like receptor 4 (TLR4) 及髓樣分化蛋白 2 (myeloid differentiation protein 2, MD-2) 組成的受體複合體作用，啟動下游訊號傳遞。LPS 刺激可活化 NF-κB、MAPK 等訊號傳遞途徑，促進腫瘤壞死因子-α (tumor necrosis factor-alpha, TNF-α)、白介素-1β (interleukin-1 beta, IL-1β) 及白介素-6 (interleukin-6, IL-6) 等促發炎細胞激素的產生，進而誘導先天免疫與發炎反應。LPS 濃度分析廣泛應用於內毒素刺激、先天免疫、巨噬細胞活化及發炎反應相關研究。
