細胞凋亡 (apoptosis) 是一種程序性細胞死亡 (programmed cell death) 形式，旨在清除組織中多餘、受損或衰老的細胞。在正常的活細胞中，帶負電荷的磷脂醯絲胺酸 (phosphatidylserine, PS) 定位於細胞膜的內側（胞質側）；然而，當細胞進入凋亡程序後，PS 會由細胞膜內層翻轉至外層，進而暴露於細胞外部環境中。

Annexin V 是一種分子量為 35–36 kDa 的鈣離子依賴性磷脂結合蛋白，對 PS 具有極高的親和力。Annexin V-AbFluor™ 488/PI Apoptosis Detection kit (KTA0002) 即是利用此特性，以 Abbkine 專有的 AbFluor™ 488 綠色螢光 (Ex/Em = 491/517 nm) 標記 Annexin V，用以精準鑑定並定量凋亡細胞。為了進一步區分細胞狀態，套組中同時結合了 Propidium Iodide (PI) (Ex/Em = 535/617 nm)。PI 是一種無法穿透完整細胞膜的核酸染劑，僅能進入細胞膜完整性受損的死細胞中標記 DNA 並產生紅色螢光。透過 AbFluor™ 488 與 PI 的雙重染色分析，研究人員可利用螢光顯微鏡或流式細胞儀 (flow cytometer) 輕易區分以下三種細胞群體：

活細胞 (Live cells)：Annexin V (-) / PI (-)
早期凋亡細胞 (Early apoptotic cells)：Annexin V (+) / PI (-)
晚期凋亡或壞死細胞 (Late apoptotic / Necrotic cells)：Annexin V (+) / PI (+)

本試劑盒所採用的 AbFluor™ 488 性能顯著優於傳統的 FITC，其螢光強度更高、極具光穩定性且不受 pH 值變動影響。本套組穩定、快速且操作簡便，是研究細胞發育、藥物誘導凋亡以及癌症生物學不可或缺的分析工具。

HeLa cells were induced with camptothecin for 24 hours and stained with Annexin V- AbFluor™ 488/PI Apoptosis Detection Kit. The cell is a late stage apoptotic/necrotic cell with both Annexin V- AbFluor™ 488 and PI staining (green membrane with red fragmented nucleus).

HeLa cells were induced with camptothecin for 24 hours and stained with Annexin V- AbFluor™ 488 Apoptosis Detection Kit. The combination of AbFluor™ 488 and propidium iodide allows for the distinction between early apoptotic cells (Annexin V- AbFluor™ 488 positive), late apoptotic and/or necrotic cells (Annexin V- AbFluor™ 488 and propidium iodide positive), and viable cells (unstained).

HeLa cells were induced with camptothecin of 4 different concentration (0、1:8000、1:4000、1:2000、1:1000) for 24 hours and stained with Annexin V- AbFluor™ 488 /PI Apoptosis Detection Kit. The combination of AbFluor™ 488 and propidium iodide allows for the distinction between early apoptotic cells (Annexin V- AbFluor™ 488 positive), late apoptotic and/or necrotic cells (Annexin V- AbFluor™ 488 and propidium iodide positive), and viable cells (unstained).
